Vipergen launches new DEL series with Lib100 and updates Series 50 to Lib058
Vipergen has synthesized and released Lib100, the first library in a new DNA-encoded library (DEL) series, while also upgrading the Series 50 collection from Lib057 to Lib058.
About Lib100
Built using well-established DEL chemistry (amide formation and reductive amination) and powered by Vipergen’s Yoctoreactor® technology, Lib100 delivers a high-fidelity library with a one-to-one match between DNA barcode and encoded small molecule. The majority of building blocks are synthesized in-house, broadening chemical diversity and enabling exploration of under-represented chemical space.
Lib100 was developed to produce molecules slightly larger than the Series 50 libraries, which we believe is suitable for targets with larger and more shallow binding pockets along with the discovery of protein-protein interaction inhibitors.
Scale: Lib100 comprises 511 million compounds.
Designed-in drug-likeness
Lib100 was engineered to balance ADME-relevant properties from the outset:
- Lipophilicity: Avg. cLogP 1.6
- Hydrogen bonding: Avg. 6.9 H-bond acceptors (HBA) and 2.5 H-bond donors (HBD)
- Rule of 5: >99% of compounds comply with Lipinski’s Ro5 and Avg. MW of 598 Da
- Three-dimensionality: Avg. Fsp3 = 0.51; >96% of compounds have Fsp3 ≥ 0.3
- Aromaticity: Avg. 2.9 aromatic rings; >98% of compounds have ≤ 4 aromatic rings
These parameters support the probability for high aqueous solubility and cell permeability while keeping compounds squarely in drug-like space
Series 50 update: Lib058
In parallel, Vipergen has refreshed the Series 50 library to Lob058, introducing additional designer building blocks and removing legacy components that did not meet current standards. Like Lib100, Lib058 is Ro5-compliant and designed to generate hits with a high probability to be drug-like and cell-permeable.
Scale: Lib058 comprises 603 million compounds.
